Function:
Aldosterone is a yellow steroid hormone (mineralocorticoid family) produced by the outer-section (zona glomerulosa) of the adrenal cortex in the adrenal gland, and acts mainly on the distal tubules and collecting ducts of the nephron, the functioning unit of the kidney, to cause the conservation of sodium, secretion of potassium, increased water retention, and increased blood pressure. The overall effect of aldosterone is to increase reabsorption of ions and water in the kidney -- increasing blood volume and, therefore, increasing blood pressure.
Location:
Steroid receptors are intracellular. The aldosterone mineralcorticoid receptor (MR) complex binds on the DNA to specific hormone response element, which leads to gene specific transcription.

some questions for the ELISA kit
We have purchased the Human Aldosterone ELISA (Cat # E0911h) and the Human Renin ELISA kits (Cat # E0889h) and have some questions.
1. In the Sample collection and storage section of the package inserts for Plasma it states “Store samples at -20°C or -80°C.” Then under the Note: 1. “Samples to be used within 5 days may be stored at 2-8°C, otherwise samples must be stored at -20°C (1 month) or -80°C (2 months) to avoid loss of bioactivity and contamination.” Can you please clarify which sample type the Note 1. refers to? Can plasma samples be stored at 2-8°C for 5 days and used in the Aldosterone and Renin ELISAs?
2. Can you please provide stability data for plasma used in your Aldosterone and Renin ELISA kits?
3. In the Aldosterone ELISA, it is a competitive assay. In the Calculation of results section, it states “Average the duplicate readings for each standard, control, and sample and subtract the average zero standard optical density.” Since it is a competitive assay this causes all of the data to be negative. Please advise how the calculations are performed. Can you provide an example?
