Heparan sulfate (HS) is a linear polysaccharide found in all animal tissues. It occurs as a proteoglycan (HSPG) in which two or three HS chains are attached in close proximity to cell surface or extracellular matrix proteins. It is in this form that HS binds to a variety of protein ligands and regulates a wide variety of biological activities, including developmental processes, angiogenesis, blood coagulation and tumour metastasis. HS has been shown to serve as cellular receptor for a number of viruses including the respiratory syncytial virus.

Many thanks for your email and the attached paper. Checking the reference I note that it refers to an old catalogue number (saying: “Extracellular matrix proteins were also quantified in supernatants using commercial ELISA kits and following the manufacturers’ instructions: heparan sulphate (E0623h,EIAab, Wuhan, Hubei, China)…”) rather than with the current catalogue number E0623Ge…
Can you confirm whether old E0623h and new E0623Ge are the same kit under different catalogue numbers?

Technical enquiry about the ELISA Kit for General Heparan sulfate
My customer has a technical enquiry about the ELISA Kit for General Heparan sulfate (Cat. E0623GE). On your protocol at the top of page number 3, you specified in bold text the following warning :
“The reconstituted Standards Detection Reagent A and B can be used only once.”
In regards to the above statement, my customer has the following two questions :
1.- Does this mean that the reconstituted standards, AND the Detection Reagent A (diluted 1:100) AND the Detection Reagent B (diluted 1:100) can only be used once?
2.- Is it possible to reconstitute all 3 reagents and store 4 degrees Celcius for a day or two if some remains for another attempt at assay?

1.The reconstituted standards, AND the Detection Reagent A (diluted 1:100) AND the Detection Reagent B (diluted 1:100) can only be used once.
2. I would recommend diluting the required amount based on the volume used for the assay.

some concerns for this kit
I have read the information provided with the kit and this raises some concerns. Please explain how you can have recombinant protein as the reference standard when the assay is for heparan sulfate. Is the assay really for HSPG? Which HSPG? If so it is not suitable for our requirements.Please provide information on cross-reactivity with heparan sulfate from other species or from differing organs.

The Human heparan sulfate ELISA is designed against the in-vitro quantitative determination of heparin sulfatein human serum, plasma, buffered solutions or cell culture medium. The assay is highly specific for Heparan Sulfate Proteoglycan. There is no evidence for cross-reactivity with other connective tissue proteins (vitronectin, fibronectin, elastin, collagen, laminin) but it does cross-react with human thrombospondin.
