Coronavirus tropism is predominantly determined by the interaction between coronavirus spike (S) proteins and their corresponding host receptors. The S protein of SARS-CoV-2 (SARS-2-S) can be cleaved into the S1 (SARS-2-S1) and S2 (SARS-2-S2) subunits, which are responsible for receptor recognition and membrane fusion. Binding of SARS-CoV-2 to ACE2 occurs via the C-terminal domain (also called the receptor-binding domain (RBD)) of SARS-2-S1. The observed incomplete inhibition mediated by ACE2 antibodies and the high neutralization potency of monoclonal antibodies (mAbs) targeting the N-terminal domain (NTD) of SARS-2-S1 suggest that SARS-CoV-2 might use other (co)receptors or other mechanisms for host cell entry.
